Update: Decided to pursue an RX350 instead of a GX460.
I test drove a GX460 at a dealership (it was brand new, under 10 miles on it). I took a 6 mile test drive, drove very conservatively, and the MPG gauge showed 9.9 MPG (I'm assuming they ran premium fuel).
The car looks great, much better than the RX350 in my opinion, but it just doesn't fit my specific purpose, and the low MPG sealed the deal.

Something is real wrong with that MPG, but you've go to do what's best for your situation.

Break in needed for the true MPG? Under 10 miles on it... Any new owners here notice a change in MPG over time?

It's probably still running the Japan gas and has had a ton of short trips, a lot of idling time and a ton of start ups. Need to run a full tank of gas in it to see real MPG's. Out of factory, to a staging lot, to a truck, off a truck to a stagunf lot, on to a ship, off a ship to a staging lot, on to a truck and off the truck to the dealers lot, no way to tell real MPG that way.

I'm supposed to get my factory ordered 2016 GX 460 in approximately 10 days but one would assume that if it was fine to run regular gas vs. premium most in this forum would be saying that. However, you could always try running a full tank of premium, record the relevant data and then run a tank of regular compare the two and post your results on this forum.
FYI, my factory order will have taken just over 3 months to get my dealer, which imho isn't all that bad at all.

I average 16-19 miles a gallon on premium. Mixed highway, city, stop & go. Lowest I have gotten is 14.5 (when I wait to long in the pickup lines for school on idle) and max is around 21.2 mpg (highway) on highway we are normally pushing 70+ when its open.
We dont lease, if I did maybe I would put regular. Since its ours, I take care of it and only use premium.
Its a great ride, we love it!!! Enjoy.
We came from a Sequoia, and I will say I miss the extra room and storage. But the drive is awesome.
What sucks is that I'm running premium and only getting (according to the computer) 14.6mpg on a tank. This is 90% city driving in southern California. I'm very conservative in the way I drive. I've been recording my fill ups manually but haven't done the math yet as I've only had two fill ups since I bought my 2010.
I will say on a quick trip to Palm Springs I recorded as high as 22MPG which was pretty much 95% highway.
