For some reason the 2015 GS 350 I have can no longer lock doors via swiping the fingers on the door handle anymore. I can still lock the door via the remote key though. Is there a setting somewhere that I need to set/check?
How it works is your car is constantly scanning for the FOB. When you walk close to it, or touch the handle it picks it up. Disabling this feature is actually a smart move. Because enabling "Access System with Elec Key" is said to make your car vulernable to relay hacks. Another smart move is to simply park inside, and let them pick an easier target.
Also, i'd also try changing the battery on the key fob. I've noticed that once the key fob battery starts to get weak, the lock/unlock features are intermittently impacted.
I have to say that there is no issue with the key fob. I can lock and unlock with the key fob. The issue is when I tried to manually lock the doors by swiping my finger on the door handle.
The key fob is needed to lock and unlock when using the keyless/entry system. If the key fob's battery is weak, i've personally had issues with sensors being able to grab the signal being sent from the fob itself. Before taking it to a shop, I would try a new fob battery to see if that helps. Otherwise if you have another spare key fob, put that one in your pocket and see if that one works to lock.

I don't 'swipe' anything - I just touch the indentation sensor on the upper part of the door handle or grab the inside portion of the door handle.

We tried touching the indentation sensor and it didn't lock the vehicle either. We tried multiple doors and it's not locking that is why I assumed it might a setting somewhere. Unlocking still works by simply grabbing the inside portion of the door handle and open.
When using Smart Access to unlock the doors - does gripping the driver door handle unlock just the driver door (which is 'Standard') or does gripping the driver door handle unlock all the doors?
When you try unlocking all doors with the key fob listen carefully at each door to determine if you hear the solenoid door actuator at each of the doors. One bad actuator can upset the door module operation.

Thanks! I will try to listen to it the next time. What got me confused is that with the key fob lock and unlock works with every door just not locking when touching the indent on the door.

If all doors lock and unlock with the key fob then all the door actuators are okay.
I suspect you'll have to use Techstream to determine if all the door handle indentation sensors are able to send a signal to the door module, and if the module is okay.
