1) If you have ML with your GS, then you can do the override which is one of the stickies up top of the 3GS section.
2) ECU/Computer wise, nothing that I've seen yet. I've heard of nitrous being used, which is a great idea. Just don't get too crazy with it. Other then that, basic bolt-ons make a LITTLE bit of power but to some it makes a difference. I'm not willing to fork out a few hundred bucks to make 10hp.

300hp from 3.5L is pretty amazing. Most 3.5L engine makes ~260hp. Don't expect to get more out of this engine by bolting on some aftermarket stuff or modifying ECU.

As the others said, there's no plug and play hp for the GS350 or any Lexus or Toyota for that matter. You can add small bolt ons like an intake or exhaust to help define the sound, but hp gains are very minimal and usually go unnoticiable.

As for watching DVD's with a ML/NAV GS, you can do so with either the old way of modding the wiring or you can opt with the latest option, the plug and play R-Spec Datasystems NAVI/TV override unit.

40+ hp to get out of the 4.3L V8 3UZ-FE is going to be very tough. Unless you have deep pockets, it's not going to be easy to get there.

It's tough because of the ECM and the parameters the V8 was set with. If you want to add power, you can go F/I, but that's when it starts costing money in terms of fuel mangement and etc.

F/I = forced induction either turbo or supercharger.

As for the nav override module, there's a huge thread discussing it here:

Not entirely true... there is a TRD supercharger for the rollas and tC

Yeah true but also for the 4Runner, Tacoma, tC, new Tundra and etc. you have to pay to play.
