Something I have looked for also and not found.
I have entered road intersections and then manually added them to navigation takes me a certain way. I would like to save "Smiths via 505" but have to manually enter the two destination. I am happy that Lexus Navigation, unlike Google, allows for multiple destinations.
One, after passing through an intermediate destination, Navigation tried to get us to turn around and return us. I guess we didn't stop so it figured we had missed it. Now we use the voice command, "Delete Next Destination" when we are getting close to an intermediate stop.

Disappointing Answer - No Multi-Destination Support
Spoke to the Nav Unit Expert at my local dealership. Multi-stop routes are not supported as address book entries.
