I own a 2007 RX350, check engine light, when I plug in my scan tool says misfire cylinder#2, that is the only code, I Replaced the Spark Plugs for #2,#4 and #6 and the PVC valve, cleared code and it keeps coming back, the Engine Idles smooth and no drivability Issues,any direction I should pursue next? I don't want to fire the parts Cannon, Thx
First time I swapped the coils, cleared code and it came back, next I put in a new coil pack, cleared code and it came back (Misfire cylinder 2 every time) I'm pretty sure my scan tool shows the short term, Long term fuel trims, I will check that and post the findings
Could be a fuel injector. Sometimes a couple tanks of Mobil Super Premium will clean it up. Sometimes a pressure cleaning will do it. Sometimes RockAuto has injectors on sale and new is the way to go.
Considering the misfire, the fuel trims do look normal. I would think, someone correct me if wrong, if a fuel injector was the problem, the trims would be different on bank 2? Tricky one...
I had this same issue on a Honda Odyssey and it turned out, after trying everything else, to be the fuel injectors. The fuel trims looked great. I adjusted the valves which the internet said was the problem. New plugs. New coil packs. New timing belt. Adjusted the valves a second time. Turned out to be the fuel injectors. I'm not saying "that's what's wrong!!!", just saying its an avenue for exploration. And RockAuto has them for $25 each so, depending on how hard they are to swap out, that might be an inexpensive test.
You mentioned the car runs smooth and fuel trims look to be in spec so your job just got harder if you already swapped the coil and spark plug.
How often does it trip the misfire code?
Could be fuel injector as already mentioned but hard to know yet. I did a seafoam intake treatment but I had a rough idle.
I get a check engine light after Refuling, but sometimes while driving, I have ruled out many things, so I Replaced the gas cap yesterday, the old felt loose to me, I believe it was the original, I'm hoping that is the reason for my misfire cylinder 2 code because when I go to clear the code my scan tool says are you sure you want to erase this emissions related code,time will tell

Ah yes, you may have more than one code.
There should be a page button on the scanner to flip to the next code or "pending code" since it sounds like you are clearing a misfire code and an emissions code(which sometimes will be pending but not throw a light).
UPDATE: After installing a new OEM Gas Cap Yesterday,courtesy of Amazon 😚
I Put over a Hundred miles on the Lexus today, even refueled the tank and No Check Engine Light
It was a Faulty GAS CAP that was introducing unmetered Air into the Intake and was causing a check engine light with a misfire cylinder #2 code 😱
Problem Solved thank goodness 👍🏻
BTW I had No other codes and No Pending Codes either
Glad it is resolved because quite honestly I was running out of things to check
In the past I am guilty of"Firing the Parts Cannon"
but now I really try to Research first and to test suspected parts and or Sensors 🤔
I read so many peoples posts about an Automotive issue and they never post an Update, I feel it's important because it may very well help out someone else who May encounter a similar issue with there Vehicle, this is my first Lexus and I Absolutely love it ❤️, wish I would have purchased a well maintained used one (as mine is)sooner,I can see and feel the build Quality 👍🏻

This doesn't make sense though. Put the old cap back and confirm #2 misfire returns?
Yeah I can't believe that a bad gas cap can cause a check engine light with a Misfire cylinder #2 either but I subscribe to a utube channel called The Car Care Nut,he works for a Toyota service dealership, I told him my issue and he said it's either my evap purge valve stuck open or my gas cap, my purge valve is working normally so I Replaced the gas cap and no more check engine light or pending codes, to answer your question, no I didn't put the old gas cap back on but I'm 💯 sure if I did the Engine light will return with the Misfire code
