I jacked up the RX300 using the front cross beam member. Then I put some cinder blocks and wood under the "round things" on both sides and lowered the car on them.
The "round things" are illustrated in the photo mercilessly stolen from LexMex and modified yet again. The green circles indicate where the car was supported.
I have read that cinder blocks are the one thing you should never use to support the weight of a car. The reason given is that they can fail suddenly, without warning; whereas wood, metal, and other materials will usually give some warning prior to failure.
I think you have the floor jack point correct. The Repair manual says to support the car from the points where you would use the spare-tire jack. Those seem to work OK, although there are probably other good points too.
If you must, then the holes should be vertical {the picture shows them being used incorrectly**. Concrete can take compressional force. The span over the holes can not support much.

Good point - should have had them rotated 90 degrees.
