Have a 16 LS460 with the stock infotainment system. I have my iPhone paired to the system.

Is there a way when I use Google Maps that the directions from Google Wench will play through the speaker system? In other words, play over whatever radio/sat is playing while I am driving?

I prefer to not have to hold my phone or look down to see the next turn. I prefer audio directions.

It cannot play over the car system radio/CD.
However, you can put on Bluetooth audio and use your phone for your music(iheart radio, tidal, spotify, YouTube, etc) while having the google map on and it will interrupt the audio and announce the turns and direction.

You can pair your phone as a "Phone" or "Audio Player". When you pair your phone as "Audio Player" it will be playing through the speakers. Sometimes I use my phone this way when watching some youtube videos, also used it with Google Maps. Just have to select Bluetooth on your stereo as a source.
