My check engine light came on. I have replaced the fuel cap but the light still stays on . I have no problems with the drive train. What should I do?

The most common cause for our cars is one of the Oxygen sensors. Get the code from your local mechanic (autozone, etc) and you will know where you stand. Lexus will most likely take you over the coals.
If you have an OBDII system, usually 95 and above, then you can go to Autozone and have them read the codes and give you a printout... then you can come back here and use the search function on the codes and see what they mean and what others have done to fix them.
If you are prior to 95, then you'll need to get a paper clip and look up some other good instructions here...as I've not had to do that. The light can be triggered by a large number of reasons. Just find out the code, then start working through the possible causes for that code with the help of very knowledgeable members from this site.
My check engine light came on again. I took it to Auto Zone and the OBDII showed PO135 which means the O2 sensor is bad. I will take it in this week. Will it hurt the car to run it with a bad O2 sensor?

Just do it before emissions inspections and drive few times to clear the codes (if emissions are applicable in your state).
absolutely it will... but let me first ROFL about 'changed gas cap and it's still there!!!'... how gullible... LOL
ok, when upstream sensor/s go out, the ECU will default to 'safer' fuel maps, thus seriously degrading the power output and decreasing mileage significantly.
when downstream/s give errors, that usually means that CATs are gone. at this point ECU still can properly adjust the FA mix, but it will not, due to emission concerns.
I thinks its possible to mess up the catalytic converter if you drive around with bad O2 sensors - and bad thing since the Cat is very expensive to replace.
