To flush my 2002 LS430 cooling system, I’m using the Prestone Flush n Fill Kit. Is Hose 1 the proper hose to cut and install the T? Is Arrow 1 a good spot to cut and install the Prestone Flush n Fill T or is this the wrong hose?

I wouldn’t touch either of those unless you have a genuine Toyota/Lexus part to replace them after you’re done.
You won’t find it at any parts store and bulk hose definitely will not suffice.
By looking at the kit the tee does have a cap and goes into inlet side of heater core. A long time ago at our dealership we recommended this and ended up replacing radiators and heater cores from being clogged by the backflush. I will never do this to any of my cars as I know what can happen.
