Default Check System Light

Out of no where while driving my 2006 Lexus IS350, a Red Check System Light came on. Pulled over, no flat tire found. Went to get air 35/37 per spec. Red Check System Light remains on. Tried push the button under steering wheel to initialize tire pressure, tuned off engine. The Red Check System Light is still on.Please help! Thank you!

check engine light on? if so take it to autozone and get the codes pulled. If no check engine light on, take it to the dealership for diagnostic.

"Check system" is the TPMS error that comes up when it isn't getting a signal from the tire sensors, not when the pressure is wrong. Often that's because somebody played with the TPMS menu and accidentally selected the second set of tires, but since your car is over 10 years old, it's equally likely it's the batteries in one of the sensors. I'd call around and find a tire store that has a TPMS tool like the Bartec that can individually scan each sensor. If your sensors are original, they are definitely running on borrowed time, and all four should be replaced.

I'm not sure what the dealers charge. Looks like a Denso TPMS sensor for the '06 IS350 from Amazon is about $35.

Don't go to the dealership. Bought used OEM ones off eBay for $80 shipped. They were already programmed for my 2008 Is 250.

I'm afraid you are mistaken about thinking that they were already programmed, XSV. Each sensor has a unique serial number that needs to be programmed into the car. I am guessing that the tire shop did this for you when they mounted them on your rims. With the Bartec tool, it only takes a a minute or so to scan all four sensors, then plug into the OBD2 connector under the dash and upload them to the TPMS ECU. (even the eBay listing you linked mentions that they need to be programmed to the car)

Personally, I think I'd rather buy new ones. The sensors have a lithium battery that is only good for about 10 years, and if you buy used ones, you don't know how much battery life they have left.

That's what I was thinking, most of those ebay listing never specify what year they came out of. You could end up replacing 2006 sensors with ones from a 2007, which likely won't buy you much time.
