I think you need to do more reseach on this car on Lexus.com rather than asking such simple questions like this. Also use the SEARCH function next time before you post.

An electronically controlled five-speed automatic transmission with manual upshift/downshift switches conveniently located on the steering wheel.

If you still don't understand the E-Shift, go to a Lexus dealer and ask to test-drive either an automatic IS300 or a GS300....the GS has the same engine-transmission combo with the same E-buttons on the steering wheel. The E-buttons are for use when the shift lever is in the "M" (manual) position. They will allow push-button shifts from 2-5 except at cold engine temperatures, which will block out 5th, and 1st is not accessable with the buttons...you must use the lever. 4th is not accressible with the lever...you must use the buttons. The computer will keep you from overreving the engine no matter what gear you select. And, yes, the tranny goes back to first at a dead stop no matter what the buttons say, and upshifts normally to the chosen gear from a dead stop
Now....does THAT help answer your questions?

Do your own research. Especially with such a simple question like that. If you were really interested in the IS, you would know what E-Shift is despite the fact you want a manual or auto.

Default E-Shift or manual?

Maybe what this guy really wants is neither.....a sequential paddle-shift manual BMW-style.
One more reason for Lexus to consider it.
