I have a 2007 RX350 with 117,000 miles and purchased it 10,000 miles ago. I have done two drain and fills of the transmission fluid and don't think it was done prior to that. Is there a transmission filter that I should be replacing as well?

There's a filter. Check in the DIY sticky or search to see DIY's. You'll need a ratcheting 10mm wrench to get under some of the frame rails that are in the way in order to drop the pan. Other than that, it's pretty straight forward. I'd use OE filter, but most parts stores have filters that are good like Wix.
