The '10 cabin feels cavernous compared to my '04. The first week I drove my 450h, I felt small. I believe the '10 RX is wider by an inch or two. Seats are a little flatter and bigger. The dash is lower and not as close. Nav screen is recessed. Lexus took away the arm rests and replaced it with a center console that could double as the black hole. There is the pass through which to me is wasted space. It is more room than the moveable console pushed all the way back. Access is limited through the sides though. The glove compartment is bigger with removable shelves. In the cargo area the wheel well is not as obtrusive, making it wider. I noticed it takes more effort to reach items close to the rear seats.
You're best bet is to go down to a local dealer and see for yourself. The 2010's do feel bigger though, i got a ride home in one when I dropped my wifes off for service and it definately feels bigger. The 2nd gen rx's feel like you're in a mid sized car when im in it, there's not a lot of room but they drive and look deadly.
The nearest value the manufacturers provide to answer your question is Passenger Volume. In this case, the 2010 is 2 cu. ft. larger on the inside.
Plus somewhere north of 300 lbs of additional curb weight, though some of that weight comes from the larger base wheel diameters and thus heavier wheels and tires.
I obtained an RX2010 loaner for a day. I like the older body style and interior better. The ride on the 2010 was a bit harsher, maybe because it's newer, and the interior console looks like it was designed by 3 different people. It does not flow well like the 2nd or 3rd gen models. I did notice however that it is much easier to see out the back window. I also felt a bit claustrophobic because of the large console stuck between the driver and passenger. I will not be purchasing this model.

So what loaner vehicle did they end up giving you?? So far I`v had; 2010 ES 350,2010 RX 350,and this last time a 2010 IS 250. I liked them all,and the IS is definitely a fun car to drive!
I pick the 2006 RX over the 2010 RX any day! My reason is simple: the interior is somewhat better design for your own comfort. Both drive about the same, although the 2010 RX has a bit better rpm.
I ended up with a 2010 ES350 as a loaner. These are still being manufactured on Japan.
There was a 2010 RX350 and I tried asking for it but alas he did not bite.
Well I had fun with the ES350, loaded but no NAV, 15,000 miles. I am sorry but the wood trim sucked. Reminds me a Chevy Malibu wodd trim. I kid you not. Just my impression.
I just traded in my es350 for an 2010 RX350 and I LOVE my new vehicle. I would strongly recommend the RX350 to anyone out there !!!!! I love the nav system most of all !!!
They give me 2010 gs350 with only 7000 miles on it for my loaner. Dang it felt really cramp inside compared to the Rx. But it was so much fun to drive. That car really accelerates!
