My 08 LS 600h does the same thing. 90% of the time it happens its on a slope

I have not been able to duplicate the problem on mine again. Grantetd I dont drive it much. It did it consistently for 3 days than that was it. So ive be keeping an eye on it. If it starts up again my plan is to film it as was suggested, than take it in.

I have had the problem intermittently with it not opening, only by the key. Then to get it to shut often, pull the emergency release in the trunk, shut it slowly to touch the lock to see if it shuts and usually do it again the latch opens so you can close the lid and hold it down to latch. It does this once in a while. I think most of these issue lie in the control module on the drivers rear wheel well. a electronics? what are you going to do!

I'm having similar problems as this. I can press the button on the fob, lower dash, or on the trunk and all it will do is make a "beep" noise and the trunk won't open. The only way to open the trunk is if I use the emergency release, or opening it with the key manually. When I open it manually, when I try to use the button to auto close, the trunk lid will go all the way down, but will not latch and pops back up a little.

I did find that some members on here removed the cable from the battery and it solved their problem. I tried doing so and it will only open and close like normal just one time from any of the three release buttons. Every time the battery cable is removed, it will only open once.

So Im stumped as it will open and close one time after every battery disconnect, but will not close shut when I manually open the trunk and use the auto close button to close it.

I did find a TSB from "Canada builds" that describe the symptoms, but the revised part number for the rear junction box (drivers rear wheel well fuse box) is the same part number as the one that is in my 07L as I removed mine to check a few days ago.

So I'll be keeping an eye on the thread hoping there is a solution.
