Upgrading stock ML subwoofer?
I did that in my 2019 Hybrid Highlander. It was easy and can't be much different.
As for the Lexus. Before you start replacing; try adding sound deadener behind the sub to see if that helps enough to where you can tolerate it. Also add some insulation behind the sub to filter out any reflections that may muddy the sound. If you're still unhappy, replace the sub, but find the highest efficiency sub you can fit, to get the most out of the existing amp. IF you still want more, then add an dedicated amp for the sub.
Keep in mind that the above work won't go to waste; it will help the new setup sound better.
