Try this:
Press the rubber button that is the door sensor. While holding it, hit the lock button on the door. Release the button. Close the door and call it a day.

Default Locking an RX while running

So I have a bad case of CRS! I want to lock my running car while I run into a store. I want to keep the AC running for the dogie. 2012 RX 350. I am sure there is a way.

Nope, not possible. You'll have to look into something like remote start.

You may come back to your car to find someone has smashed the window to rescue the dog from the apparent heat inside the car.
A policeman lost his K-9 officer because the A/C failed while the dog was in the car. Sad story.

Open the door, press on the lock sensor, while holding the lock sensor in place, press lock on the door, release the lock sensor and close the door.

No, the vehicle will not auto-lock when the smart key is out of range.

That explains why I could not lock my RX 350 the other day. I thought I had my key but I did not. My wife was in the car with hers and the car started when I pushed the button. (Still not realizing I did not have my key) We got to a restaurant and left my wife at the door and I then parked, I got out but could not lock the car with button on the door handle nor could I lock it with the button on the arm rest inside. Then I realized I didn't have my key and looked all over thinking I dropped inside. Finally gave up and went inside the restaurant got my wife's key and locked the car.

At least you were at the same place instead of miles apart with the engine turned off.

No kidding! In this case we had driven from the Northwest suburbs of Chicago to Milwaukee. Any number of odd things could of happened. When I started the car in the garage, she hopped out because she forgot something, if she had decided to switch purses, we would have been stranded.

I'm sure you would have noticed a flashing exclamation point in the middle of the cluster, prompting for attention. That, and the 3 beeps it makes when any door is closed, plus a text message stating the key is missing.

Comes in a Lexus flavor as well.

Can be done

Open the door, lock using interior door buttons and close the door. Leave the window open slightly obviously.

Take the fob with you, itll beep 3 times but thats it, car will remain running and no beeping.

When I travel with my dogs in the summer and stop at the rest area to use the restroom, I remove the key from inside the fob, place the fob at the bottom of the console, make sure the air is on while the car is running and lock the door with the manual key. Never had any problems. I also make sure to put the parking brake on just to be safe
