The RX 330 is coming with a 5 speed automatic transmission. I needed a Lexus loaner car recently and I had the chance to drive the ES 300 with a 5 speed automatic. If the RX 330 tranny is anything like the ES, it will be great. While it took me an hour to get used to the 5 speed, instead of my RX 300 4 speed, the shifting was smooth as silk.

I had the opportunity to test drive the sequential mode shift on a 4x4 RX330 w/ "18 wheels in the new 5 speed automatic and it was, what I felt, to be the most fun driving Lexus so far (that I could afford). I can't wait to experience the off-road test drive in Malibu on the 19th. Usually, Lexus has them on asphalt. This is the first time we will be driving all the LUV's at the same time and us employees are just as excited as you guys are!

Let's see how accurately I can describe this: It's kind of like having an e-shift, but instead of on the steering wheel, it's down on the shifter. So there you are driving, then there's a +/- sign next to "D" mode. You just move it either way, depending on whether you want to shift up or down. It was cool because I sometimes currently shift anyway on my automatic ES300 and this just seemed to have the best of both worlds. 

Pole Position
If you want to experience the fun of having the "+" and "-" shift, go and test drive the Mazda6 V6 with the sport package (besides Mazda is giving away $35 or a leather backpack for just test driving it). I had 100 times more fun using that on the Mazda6 than the e-shifts on my GS400.
