Knock sensor
I have a 2002 rx300 AWD. My transmission will only shift into overdrive on initial startup but won't go into overdrive after second stop. I have to turn the ignition off and then start the engine again for it to go into overdrive. Does the knock sensor codes have anything to do with this problem. Anyone ever experienced anything like this? I also replaced the abs speed sensor. Looking forward to any help.

Yes, the knock sensor is the problem! It will prevent the car from going into overdrive. Fix the knock sensor, and you will have overdrive.
There is a DIY on here for replacing the knock sensor.

If you're going to have a shop do it, have them take a look at the wire harnesses also. Also, depending on your total ODO miles, you could save some labor $$ and get he timing belt / water pump serviced while they're in there. Get a quote and compare,
