I can't pinpoint what's going on, so I thought posting to the group may have some ideas.

RX300 w/100K mi. Car runs great.

Whenever I put it into gear, or accelerate suddenly, I hear a faint knocking sound coming from underneath the car. I haven't been able to get it up on a lift and look yet. Anyone expereince something similar? Perhaps a muffler got loose or a common thig on the RX300?

It's your Transmission mounts. Pretty easy to replace. There should be a DIY on this website, but if there isn't, you could make one! They are not that expensive.

I know nothing about transmission mounts...do these usually just wear out over time? Is it soemthing that needs to be addresse ASAP or can I drive safely for a a while.
