2000 Lexus RX300 cranks but won't start

Hesitated we managed to get it off the road shut it off would start for a second and then die out wanted to run with starting fluid replaced the fuel pump still won't start help please

Check presence of regular spark, by pulling out #2 plug [just because it is convenient]

imobilizer will kill the spark.
