This isn't a Tesla. Your LS wouldn't have been allowed to leave the factory if it wasn't 100% perfect from top to bottom. From what I've read, the only thing to make sure is done is the software update for the transmission to fix some low-speed jerkiness that some have experienced, but otherwise, you should be good to go.

Big congrats on your first Lexus! You made an absolutely excellent choice!

As Signdetres said in another post, the Lexus brand stands behind its cars, especially the flagship LS500. I'm driving my 3rd LS (from the LS400, to LS460, to the latest LS500), and every time I took delivery of my new LS, I haven't had to check anything at all but driven the car off the lot. Enjoy your new LS!

When I picked up mine, I made sure it was a sunny day so I could see scratches and and any damage to the exterior and interior. Make sure also that all the items stated on the window sticker are in fact in the car and also the engine clips as well. For mine the battery was dead and they needed to jump it and forgot to put the clips back in. Mine was also missing the all weather mats as the car came with it.
