I typically use the Redline MTL or MT-90. I've used this on my Civic and Accord and now on the IS. The owner's manual states 1.9 quarts. And you'll have to get a hand pump to fill it up. The fill port is high up on the bell housing and you won't be able to pour anything in.

I went to Harbor Freight and bought a cheap hand pump to do the job ~$7.

Online from Summit Racing. Usually arrives in 2 days to Cali. If you order it tomorrow you should have it by Wednesday.

I think maybe O'Reilley's has it too. Check Autozone as well. One of those 2 stores carries it. Or even Napa.

Best transmission fluid I ever used is AMSOIL. I use in my Evo 9 trany and transfer cases . I also use it in the lexus is250 differentials. When I had redline shifts on the Evo used to feel knocky during cold weather switch to AMSOIL and never felt smoother just my 2 cents.

I would be careful running fluids like MTL and MT-90 in your IS250 with the 6MT, as these are GL-4 gear oils. While this may not be too big of an issue (I'm not sure of the exact formulas obviously), there is a possibility that it could cause issues later on down the road. Redline's 75W90 NS is a GL-5 gear oil that is probably better suited to the RA62, also AMSOIL's Severe Gear 75W-90 is also a GL-5 gear oil.

Originally Lexus spec'ed the RA62 in the IS250 for GL-4 or GL-5 gear oil, however after mid-2006 they changed the transmission and updated the spec to GL-5 only. Only IS250's produced before June of 2006 are spec'ed to use GL-4/GL-5, unless they've had their transmissions replaced since then, in which case Lexus says to only use GL-5.

Quote:
SPECIFICATION:

RA62 (6-speed) Manual Transmission Oil Type Requirements

For IS 250 vehicles that have the RA62 (6-speed) manual transmission with VINs produced AFTER the
Production Change Effective VIN shown above, use ONLY manual transmission gear oil meeting API GL-5
designation.

CAUTION:
Using manual transmission gear oil other than API GL-5 may cause occurrence of gear noise
and damage to the transmission.

For vehicles that have the RA62 (6-speed) manual transmission produced BEFORE the Production Change
Effective VIN and have had the manual transmission replaced with a supply part, ONLY use API GL-5.

Built BEFORE Production Change Effective VIN: Use API GL-5 or API GL-4
Built AFTER Production Change Effective VIN: Use API GL-5
With Replacement Transmission Installed: Use API GL-5

PRODUCTION CHANGE INFORMATION:
IS250 Production Change Effective VIN: JTHBK262#62018969 (Production 06/2006)
As for my car, I use the original factory Toyota Gear Oil SUPER 75W90 GL-5 (08885-02106). I've had the best luck running factory fluids in most cars I've had. I've also run Redline and Royal Purple and had good luck as well, but even then on a couple of cars when I switched back to the factory stuff it was better shifting. Different cars and transmissions may vary of course.

Use GL-5.

08/05-06/06 = GL-4 or GL-5
06/06-on = GL-5 only

GL-5 is best, it applies to everything, you cannot go wrong with GL-5.
