Default remote/manual lock not working

Anyone else have a problem with remote and manual lock(pressing the black button on the drivers door) not working? My battery was low so I switched to my spare. When I tried to start the car, it asked for the key to be by the start button. Maybe the battery is low but would that cause an issue with locking the door manually also?

I have a 2008 IS250...same issue. I drove to work, parked it. Got out and pressed the black button to lock the door and the car wouldn't lock. Nothing happened at all. Then I tried the remote...nothing, unresponsive to all remote commands. This has happened before, just the remote. I replaced both remote batteries and it hasn't changed at all. What would cause this. I read the post before, "Both low on battery" do you mean the car battery as well?? Lexus checked that out too and said the battery was ok.

Did you check to see there is any fuse for power door lock? Maybe the fuse is blown
