Car not beeping
Double check all the usualls, make sure the car is in park, all the doors and trunk are closed, etc...
If it still doesn't beep, you might have a faulty door lock actuator. Check and see if one of the door locks is not locking and that might be your problem...

If it is not beeping on the lock/unlock and trunk open, there is a high possibility that the piezo beeper has failed. If this is the case, it should be brought in to have it replaced.

i was doing a project on the car today and accidentally blew a fuse. when i went to go find which one blew, i ended up reading what all the different fuses are used for. n if i remember correctly there was a fuse that included the beeping as a function. so check that fuse maybe. thats if u have time. haha or else like edgeucated said, take it to the dealer!
Was it beeping before when you bought it or it stopped all of a sudden?
If you bought it like that (never beeped), then the previous owner may have the beeps disabled through LPS (Lexus Personalized Setting). This allows the owner to re-program his/her car to the way he/she want. The beeping is one of the option in the LPS sheet.
