I'm going to be waking up my GS in a few weeks after it's winter slumber. I came across clear flood mode and am wondering if my car supports it. I would like to prime the oil system before it fires if possible.

I've never heard of Flood Start or Clear Flood mode on Lexus.
If you are really concerned, pull the fuel pump fuse or relay, crank it for few seconds, install the fuse or relay and start it up. Not sure if GS 430 has a fuel pump fuse but it has a relay.
However I wouldn't worry about priming it after being stored for few months. I have more cars than I'd like to admit, some are sitting in the storage for 8-9 months on battery tender, and when I wanna drive them, I just start them up and let them get to operating temp. For a peace of mind, change the oil before starting it.

True enough. I've started and resurrected a few engines, the oldest hadn't run in 40 years.
Usually I jump on the starter so it gets a few turns in before the fuel makes it in.
I came across this in the manual, page 469. I don't get the cranking hold but it sounds like it.
I'll give this a try another time but not on this start.
If the engine will not start, your engine may be flooded
because of repeated cranking.
If this happens, push the engine switch while the brake pedal
is depressed with your left foot and the accelerator pedal is
depressed with your right foot. Then the cranking hold function
stops automatically, and you can try starting the engine with
your foot off the accelerator pedal.
You bet. Just before I parked her, oil change and filter, Mobil 1 high mileage extended range. I'm using a Lexus oil filter and I pick up a new crush washer as well. I still change every 5-8,000 kms, plus stabil with a full tank of Chevron 94 octane. I have a NOCO battery tender on it as well.
