My question is if i take out the throttle body to clean it will i have to reprogram it?

Don't think you have to "reprogram" it of any sort. IIRC, I've read up on spark plug changes for the IS250 which require it to be removed and reinstalled afterwards with no mention of reprogramming like you describe. If you are going to clean it, I think you have to be careful not to manually move the throttle body plate.

Just be careful when cleaning the inside on either side of the butterfly. These electronic motor'd TBs now are sensitive and have plastic gears.

Disconnect your battery before you take it off.

I actually removed it so that I don't get carbon inside the manifold/engine. And boy, there was excessive build-up. I don't know if my Intake was a reason for that.
