Definitely find an F-Sport Quickshifter if you can. They're unfortunately discontinued now and quite difficult to find, but if you can, it's worth it.
The transmission isn't the greatest. The 2009+ updates made the transmission a bit better, as did the TSB update, but unfortunately you're past warranty on that stuff. A few people have experimented with different clutch setups, etc., but unless you're having issues I would just leave it alone.
Sway bars are a popular and nice upgrade, as can be a different suspension setup, but that can depend on how you think it feels.
Either way, I loved mine.

I've put 45,000 miles (60,000 kilometers) on my 6MT.
By far the best car I have ever owned while at the same time having the worst tranny/clutch-flywheel setups ever encountered.
Its shockingly bad.
That being said, I'll keep it until the carbon build-up kills it and get something else.

I don't have many complaints with the transmission itself, though the shifters on the 06-08 did leave more than a bit to be desired. A quickshifter on an 09+ RA6x makes it feel pretty decently. I have the further-updated TSB transmission as well, which didn't really change the feel from the original transmission in my 2011. The factory clutch setup on the other hand is surprisingly bad in the IS250. Now that I have changed out to a single mass flywheel with a sprung disc, heavier pressure plate, and no hydraulic accumulator I've finally found that a good feeling clutch can be had in the 2IS. In fact, and I wasn't expecting it to, the new clutch setup even got rid of some of the unpleasant sounds the transmission made when at idle, so that was a nice surprise.
