So who has a K&N filter in their is250/350? And do you think it is better or have any advantages over the oem filters?

I am looking into getting one for my 2011 is250.....no mods.

I added a K&N when I put the Joe-Z on and there was a boost but I think more from the intake tube rather the filter. If you keep everything OEM around the filter there shouldn't be much gain, if any, since you wont be changing the air flow pattern. The aftermarket intake create a smoother airflow in which case theoretically you would need an improved filter to avoid that being a restriction.
