All in all the ISX50s are pretty solid cars with minimal issues...but with any used vehicle, I would highly recommend having the car looked over by a mechanic before purchasing. Could be a number of things creating the noise...

Definitely get the car checked out by your mechanic, even though lwxus and toyota hold a great track record on reliability, you never know what/how the previous owner may have treated the car.
Start up should not make any sound if you're wondering
Could you describe the noise?
Also what's the price the dealer is asking?
If it's too good to be true it usually is.

Check if it has had all the recalls done.
My car had the rattle noise on cold startups when I first got it but an oil change helped a bit. Then when I got the recalls done, it has been quiet ever since (about a year).

Take it to Lexus and see if they will do the recall on it. If so, buy the car and then have Lexus replace a bunch of parts while they have the engine down like the timing chain, spark plugs, ect.... You will basically have a new engine on the cheap!

My IS350 started up a little loud when I bought it, I got it checked out my a lexus mech. everything checked out fine. my guess is because it wasnt started for awhile.

Get the VIN and call Lexus, 1-800-25-LEXUS. Ask them if the valve train recall is still outstanding on that car. If so, I think you are okay, if not I would walk away.
