TPMS Replacements
That happened to me at first for my winter tires when I just had bought mine, and what they realized is that the parts guy gave them a 3Gen TMPS, when they switched to the proper one it worked, But of course they had lost time to remove the tires and re-balance the wheels because of the parts guy mistake.
For people reading this page and don't know this, 4th Gen RX is using 433 MHz TPMS while the prior generations where using 315 MHz TPMS.
Those are the two standards used throughout the industry for the majority of brands. So any generic TMPS at 433 MHz will do! No need to be Toyota nor Lexus.

I bought a nice set of aftermarket wheels for my RX350. I purchased Lexus TPMS sensors upon recommendation from my mechanic to insure they would perform correctly. Ironically, they didn't work. No one, including Lexus, could program them. Lexus gave up and refunded me. I haven't decided whether to try another brand or give Lexus another chance. It's beyond discouraging that Lexus could not make their own brand of TPMS sensors work.
The sensors are good for about 10 years. I replaced the sensors on my 05’ tundra two years ago when the low tire pressures light would not turn off. Costco sells the sensors for for a bit less ,but not much $300. The tire tech at Costco gave me the info and I later researched it and all was true..
Yesterday, I had these installed in my winter tires:
(Schrader 33500 EZ-Sensor) by the local Kaltire tire shop. The process went very smoothly (took me on time, process took an hour as indicated, wheel nuts torqued on the way out, tires were not dismounted so no balancing required). The first thing I noticed was that the tire pressure indicator was gone on startup. I called up the tire pressure info screen and it was blank all around. I drove about 5 km (3 miles) and suddenly all the values popped right up. So far, so good. Cost was $318 CAD ($235 USD). Happy.

I have aftermarket wheels on my RX with aftermarket TPMS valves. Lexus tell me they can't program the TPMS to my car and that I must buy Lexus ones for $500+. Is this true?

I myself am in the shopping phase for a 2021 or 2022 RX350 F-Sport, and hopefully we will be owners soon. I am proactively looking into wheels for winter tires and seem to find several different part numbers for Lexus TPMS sensors. 42607-48010, 42607-53020, 42607-06090 are the three numbers I see. I guess I will have to read the sensor part number on the vehicle we purchase in order to know which one to get?
