2010 Rx350
The wife recently purchased a 2010. Two things I learned here-
For vehicles equipped with a tow prep package the rubber oil cooler line must be replaced with all metal else risk line failure and engine damage.
Timing cover leak. To fix it properly is generally too costly requiring engine removal. Most guys here say they either live with a small leak or apply sealer to the outside.
I haven't read many chronic issues beyond wear and tear. Mostly abuse/neglect. Pre-purchase inspection is wise. Wife recently purchased a 2010 107k miles. Upon inspection the mechanic said he could find nothing in need of repair/replacement other than hood supports. We later had the transmission fluid changed. It's not uncommon to find these older vehicles with original transmission fluid.
ps. Nav software can't be updated to current.

Find one with oil cooler lines replaced with metal ones
Find one with previous transmission service history
Find one with previous coolant replacement service history
Sparkplugs should have been changed or you can add that substantial expense to purchase price. Look for tow package for additional external transmission cooling which gave lower transmission running temperatures during life of car and potentially longer transmission life, if it's there the external transmission cooler is located behind driver side front lower bumper.
