2010 Rx350 drive cycle?
So either the battery was disconnected or some one used the obdII reader to clear codes.
It involves number of starts and turning off.
Cruising with speed over 45mph for some time.
If you are infrequent driver maybe a week or if you drive frequently 3 days.
The inspection station should have charged for his time and should have given you a fail slip which gives you a waiver if you are stopped with expired tags [usually 15 days ... see the slip]. If the tech did not charge you then you may not have the slip.

As an owner of the vehicle, a fail [we are not sure if OP has real problem], essentially means additional pollutants for all of us [lets leave that aside we are in SUV forum and not riding bicycles], but most importantly we are slowly draining our wallets, if we do not address the issue(s).
Texas counties [where the emission test is required], offer assistance in repairing emission issues.

I recently disconnected my battery to perform some work on my 2015 RX350 and have to get it to emission ready by the end of next month. So I am just driving my RX350 as normal which include some varied driving conditions for over 60 miles on a two way commute to work and back. General that gets my RX350 back to emission ready on my scan tool after a week or so.
I do not know any quicker methods. I just drive my cars for a week for over 20 miles a day to get them emission ready. Always work for me unless there is some mechanical or electrical issue that needs to be sorted out that is emission related.

Also, different states have different emissions inspection requirements. Here in North Carolina, your vehicle will pass inspection as long as no more than one readiness monitor has not completed. Does not matter which one. So always know your state's rules before you go.
