Noisy wind sound on 2004 RX 330

Check your roof racks for play and sunroof seals. Next maybe check your underbody panels are all on correctly.

I am experiencing the same thing coming from the driver side backseat window. I thought the weather stripping had been torn, however, upon closer inspection, I noticed that there is a missing fastener that secures the weather stripping trim to the door. I don't know if that is an item I can pick up at any auto store, or if I'll have to swing by the dealership, but either way, it will get fixed! The noise is really noticeable at higher speeds.

brought our 2004 Lexus RX330 to dealership yesterday and they were able to install the the missing clip (door panel plastic rivets) on both front doors. did not hear the noisy wind sound yet cause i cannot drive that fast due to Los Angeles freeway traffic. I'll try again this weekend hoping for lighter traffic and let you know.

Not 100% solved but happen with the results for a 14 years old suv fix

At one shop mechanic did a test drive to listen to sounds from suspension and recommended replacing of 3 wind seals on driver's door/side. Yes, on highway wind sound was disturbing me, so I looked for these 3 seals. They are pricy for 12+ y/o car, so I decided to start with the replacement of one seal - LEXUS 04-09 RX330 RX350 RX400h LEFT DRIVER DOOR SEAL 67862-48030. Now, after 6 months, I can say that it helped.
PS: Don't expect a positive effect within the first several days/week (it can be even noisier) - new rubber part needs some time for "adjusting" for door/car body frame.
