In the RX300, you want to shoot it through the PCV valve area. There is a hose in a location similar to what this guy shows on the left side as show in my photo below (what I am holding in my hand you want to pull it off the PCV valve below).

You pull it from this point on the PCV valve. You then need to attach a funnel on this end to it to pour the Seafoam (or whatever else) in.

I should apologize as I have a hunch why. The 2001-03 RX300s have a different PCV valve setup that is a metal/brass. There was a photo a while back that I could not find. In both cases, all you have to do is get the hose off of the PCV valve, the PCV valve can stay put.

As a result of this process, your intake manifold can get yucky and disgusting with oily crud.
Seafoam is a product that can help remove this kind of stuff like carb cleaner, though more efficient. Seafoam has some other good properties in the oil and gas, but the use through the intake mainfold is the main use of it.
