We have a 2019 Premium and also had a very bothersome thump that we ended up taking to a Toyota dealership for diagnosis and repair. After too extensive-to-recant diagnosis that involved removing rear bumper, etc., it appears it was the latch below the mechanism that shuts/locks the rear door. We removed that and the noise was gone. I wasn't comfortable leaving that off, knowing Lexus didn't spend the engineering & money if it didn't serve a purpose, so added some very thin strips to the anchor (on the door frame) and reattached the striker part and put pieces of felt underneath it before screwing it back tight. Not very technical,but like i said, seems to be working. For now. And yes, it was a moderately expensive discovery due to the time required.
