Emergency Brake Won't Catch
The ratchet is integrated into the handle. It's possible that something fell in there and is preventing it from catching.
Replacing it isn't horribly expensive - the part is $150-$200. But of course you'll need to take the center console apart, and possibly remove the driver's seat for access.

When I got my 2004 gx470 the handle emergency brake was very sticky hard to release and hard to lock. I just lifted up the console wood panels and found that prior owner spilled soda or something maybe juice and it was all over the emergency brake mechanism . I cleaned ad lubricated the ratchets and the joints and problem solved
Maybe it can be cleaned but worst case you'll have to replace the entire handle assembly as mentioned above. Sometimes those ratcheting mechanism aren't serviceable internal to the lever assembly.
