You shouldn't be buying refrigerant if you don't know how to start troubleshooting the problem, I would start with making sure your wife had it turned on properly.

Never lol, no offence meant.
Unless you have experience working on ac you should just take it to a shop, experimenting with cans of refrigerant can be dangerous.

Do you have access to a scan tool (that can access more than just engine)? It'd probably tell you if one of the switches (high or low) was tripped.

You bought the can with a gauge or just a can off the shelf?

Because the one with a gauge, sure, you can put enough in there to make it work again if it leaked... but you don't always want to do that as if you don't know where it leaked from you may just be pissing in the wind.

There is a sight glass under the radiator cover trim under the front hood for the high pressure side. You may see some bubbles there when the AC is trying to run. I'd take a look and see if you see anything through the glass when you have it turned on too.

You go messing with that Freon without experience and a gauges you are asking for extra problems and trouble especially if you are not experienced.

Here is a common issue all of a sudden a/c not working with Toy/Lex. The Air Conditioning Compressor Flow Sensor switch. I gave a letter to a long time Lexus service Director to talk to Lexus about. On this problem the Denso compressor is still good. It's just the dumb flow sensor switch won't kick in the compressor to start working. This may or may not be the problem?

A quick check is to disconnect the battery with the a/c off while the car engine is cold, for about 10 mins. Reconnect the battery then cut the a/c on. You can also get a flash light and look down at your compressor to see if the clutch is turning or not, I think. You are trying to get the sensor to switch on again. I get mixed up from many different Lexus model and engines I have had before. If it does not kick on the 1st time you may want to do it again once or twice over to see. A shop can also see the milliamps =/- reading to tell if the Sensor is reading within it's specs.

This may not be the problem but, most time when all of a sudden it stops working it is. You can also google Toy/Lex Air Conditioning Compressor Flow Sensor switch problems. Its also not such a easy repair IIRC. It would save the cost of a whole new compressor though.

Hook it up, take a picture of the pressure without the AC demanded, then with the AC demanded, and post it them.

I just had an issue with this in my 15 GS350. Do this, with the A/C switched on while the engine is running and by making sure the clutch on the compressor is engaged. Hook up just the guage with the can not attached and see where the low sides system pressure is at currently at. If low or at "Add Refrigerant" level, then follow instructions on can to add freon. If low side pressure is too high, squeeze trigger and bleed off until pressure from low side is back into optimal levels. I released to much pressure when i did it the first time but had a brand new can like yourself so i just filled it back up and boom, blows ice cube temp air out now. Some of the processes i just mentioned above are not legally recommended, but they worked for me 👍🏻.
