Hello,

I am a new member, just bought a 01 GS300 2 weeks ago (only 92k). I am excited to be a GS300 owner.

My passenger door lock will only open if i manually flip it. The door lock will not respond to the key.

any idea what the problem is?

Welcome to CL. Door lock problem is likely the actuator as mentioned above. I had the exact problem when I bought my GS. I replaced the driver door lock actuator which I bought from TOYOLEX PARTS CONNECTION on eBay. Good luck with your car.
