Where is it on the 06 ES330? I took a quick look this late afternoon but couldn't find it.

On my 97 ES300, there are two nipples sticking out from the block.

 

It's pretty much the same deal for the 3MZ-FE.

Update...did my coolant change on the weekend.
I was able to take a better look while it was still bright outside.

The front drain **** is right beside the oil filter / exhaust manifold.
The rear drain **** is right behind the passenger axle bearing....it is not easy to see.
