Wheel polish
I know this isn't specific to the RX series but I'm not too familiar with the Lexus brand and which models may have the same wheels.
I have a 2006 RX400h with a dark metal aluminum wheel - what you might call gunmetal or graphite colored. It has some spotting on the wheels that I can't get rid of with cleaning. I tried Mothers Mag and Aluminum polish but it didn't turn black when I applied it. The directions said that if it doesn't turn black to try something else. I quickly tried chrome polish (knowing that it's not chrome, but thinking it was worth a shot) and no luck either.
Has anyone else had this problem or know how to get rid of these dark spots?
Thanks!

Someone correct me if I am wrong, but I think most modern wheels have a clear coat. Even aftermarket wheels that are polished are clear coated to prevent spotting. You might want to try 3M rubbing compound. It may mork.

It does look like it has a clear coat, which I would think make it easier to clean. I can't "feel" any raised or textured spot and I hope it's not something under the clear coat. Is this rubbing compound like something you would use before waxing the paint? I have something similar from Turtle Wax.

the reason it didn't turn black is because you weren't applying on bare metal. Your wheels must have a clear. Like the others said, follow the steps of 1)claybar 2)3m rubbing compound, 3)wax for a protecting finish.
If the spots don't disappear then my best assumption is that some acidic chemical got on your wheels and ate through the clear.
