Mechanically? Not really. You will hear a bit of engine ticking from the direct injection.
1. Read about wind noise. The modification with the 2013+ fins seems to be uniformly "it's better or about the same." No one has ever had worse outcomes (more noise after the fix).
It seems like the 2010+ LS460 has largely been problem free (some glitches with 07-09 cars have been reported here on Club Lexus).
2. 0W-20 engine oil should be used.

Could be...but it also could be control arm bushings, which are a common problem on these vehicles. This used to be a very expensive fix for the LS, but you can now get bushings on Ebay for very little money.
