Did my first seafoam on my '91 LS400
Just used it in fuel first......didn't notice a difference but I'm sure it's doing a good job of cleaning the injectors.
Then I used it in the crankcase, drove the car for 2 days, got an oil change and WHAMMO. Better throttle response, better power, feels smoother, and sounds smoother. I am very happy with the results.
Next I have to put it in the brake booster line, although I'm not at all mechanical so I'm a little worried about taking a house out and putting back together.

I've always been paranoid about using combustion chamber cleaners like this on high-mileage engines because a lot of times the carbon buildup is actually holding things together and preventing blow-by. How many miles are on your LS400? This is definitely promising though, I have yet to see someone say anything bad about the SeaFoam stuff.

I ran a tank through my 95 last week and noticed a smoother idle, response and acceleration. I have 175k miles and the next time I change my oil I will add it to the crank case. I did see that you could run it through the PS and AT systems but I won't do this since I have changed the fluids regularly. It says it is 100% safe though so I look forward to your replies on how it worked.

I would only drive for 10 miles it evaporates once the engine gets to normal operation temp. So running it longer is a waste as well as it has just broken down the properties that make oil good as well as the sludge.
Pour in the engine cold and drive to get a coffee and come back then change your oil and beverage to a beer and you are done.
Leaving it longer is not going to clean the engine better after the first 10 minutes.

I hope this doesn't sound like a dumb question. Do I have the car running when I do this?

Should I get an oil change after this is done?
Change spark plugs?
