How much bigger is the 350 chassis as opposed to the 250?
I find a lot of times I'll mix them up from a far and even as I'm nearing them until I confirm 100% by the badges, but close up you can tell the 350's wider set than the 250.
Are they wider and slightly longer? Or am I tripping and it's a considerable difference?
Identical. 350 apart from a badge, (visually) has bigger brakes. Inside it has the 'AUTO' on the Recirculate and Draw Outside Air whereas the 250 does not. Some 250's do have smaller 17" wheels

You literally cannot tell a 250 and 350 apart from the exterior except for the badge. The chassis is also virtually identical.

This is for a 3IS and directly from the Lexus website. Scroll down to Dimensions. As with the 2IS, the 3IS has the same dimension for all models.
The IS F is wider in the front, the rear is the same for the IS250, IS350 and IS F. The main thing that can cause anything to look smaller is the AWD's (of which are more IS250 than not) have narrower and shorter tires, but the vehicle's body is identical.
