What year is your car?
Is it a GS 350?

I believe all you have to do is flip up the small door above the hazard button, press in on the old SD card to release it, pull it out, insert new SD card until it locks into place, and close the small door.

I believe it started MY2015+. It would be where the Mark Levinson label is.

The install notes says for the Power to be OFF, and as you pull out the SD Card check the orientation of the card and be sure to reinstall the new card the same way.
If it isn't working check to see if the card can be inserted a different way.
FYI- I checked mine and the Label is Up and the contacts are down and go in first.

Good Luck

Instructions say "ACC Power" so I take it means Main Power is off.

That is what I understand from what I have read on the Lexus website for Navigation Update.
