If you do a YouTube search you will find several videos on how to remove a fender in general.
I did not find any that are specific to a Lexus, but the steps should be similar, and at least will give you an idea
Here is an example of a video I found that may help you out a bit.

These are the instructions as detailed in my Haynes Repair Manual.
So basically, there are fasteners/bolts underneath the front bumper/fenderwell, all the bolts, screws, and clips inside of the fenderwell/fender liner, 2 bolts in the door jamb, and 3 bolts along the top of the fender. It may vary slightly since the manual is (mostly) for a 97-01 Toyota Camry that shares the same chassis.
1. Raise the vehicle, support it securely on jackstands and remove the front wheel.
2. Remove the front parking/turn signal light.
3. Detach the inner fenderwell screws and clips, then remove the inner fenderwell and mud shield.
4. Open the front door, and remove the upper fender-to-body bolt.
5. Remove the lower fender-to-body bolt.
6. Remove the remaining fender mounting bolts.
7. Lift off the fender. It's a good idea to have an assistant support the fender while it's being moved away from the vehicle to prevent damage to the surrounding body panels.
8. Installation is the reverse of removal. Check the alignment of the fender to the hood and front edge of the door before final tightening of fender fasteners.
