Easy aftermarket radio replacement option!
Well, easier at least....
I just got done with a day's worth of install on my 05 RX330... I installed a Crimestopper RS-4 remote start and Kenwood DNX5120 DVD/Navigation headunit with the KCA-BT200 Bluetooth module, KCA-XM100V XM module and PAC SWI-Jack steering wheel interface.
I ordered the double-DIN radio bezel from ebay to mount the head unit with and I must say, the shipping was VERY fast on it. Mounting the radio took a little creativity, but it wasn't bad. The Kenwood trim ring doesn't fill the gaps on the side of the radio, so I modified the pieces that came with the dash bezel and hot glued them to the radio once everything was bolted together. It's not 100% perfect, but fine for anything other than a show car.
NOW FOR THE BEST PART!!
The saving grace for the install was the Metra TYTO-01 wiring harness. It's not labeled for Lexus, but rather for Toyota's with the JBL amplified system. IT IS PLUG AND PLAY FOR THE RX330 WITH STANDARD AUDIO!!! I can't vouch for any other models, but it worked perfectly on my 2005. You plug it into the 20 pin harness from the car, connect +12V, ACC, GND and REM to the aftermarket harness, then plug the RCA's from the Metra piece into the HU. You can now retain your stock amp and speakers, and it has a gain adjustment on it so you can tune it to your HU's pre-outs.
Forgive me if everyone is already aware of this harness, but I sure couldn't find any definitive info on it anywhere on the web.
On a side note, don't try to use an add-on remote start system that utilizes the OEM Lexus remotes.... it will work, but once the car is started remotely, you can't use the Lexus remote to unlock the doors. You have to use the key in the door to get in... I had to go back and do some rewiring and use the remotes that came with the aftermarket system to get it working properly. That added another hour or so to the job...

So you aren't using the ad-box1? I already bought that, going to order the steering wheel controls adapter and the tyto-1. Then get my install started as soon as that gets in.
