Did you just notice the ride is rough or it has steadily gotten worse over time?
Start by having the suspension examined and also take a ride in a loaner/friend's vehicle.

I have the same year also and thought the ride was too stiff for my liking . First off the dealer put on new Firestone tires before I bought It .
They were more like truck tires road hard over bumps . I put on Bridgestone Ecopia tires . That helped a lot, smoother and quieter .
Keep the pressure at what the door jamb sticker says I think its 32 . I bought a good tire gauge and take care of that myself . The tire shops tend to
over inflate them in my opinion . That still wasn't giving me the Lexus ride I wanted . I also put 100 lbs of sand in the back that helped a little but got tired of leaking bags big mess . So I bought new rear OEM shocks the price was good the ride was
still harsh . I was about to sell the RX when someone on this forum talked about KYB brand shocks . So I replaced my 2 month old shocks and put
on KYB . There about 65.00 apiece. Instant ride change, softened the ride just enough to take that harness out of hitting small bumps cracks in the road etc .
Since I am never satisfied with things . I put KYB struts on the front also mine has 100000 miles so I figured it was time . Now It rides like I think a Lexus should ride . Now I don't think about the ride anymore .I just enjoy driving it . Well when I take it from my wife that is .

I agree with Barry2522. I too felt the ride in our 2011 RX 350 was too harsh for what we paid for the car, so based on what someone said on this forum, I bought two KYB rear shocks from Rock Auto (sorry, I don't have the P/N) You can look it up on the Rock Auto web site. Anyway, It should have been a simple job, but I couldn't get the bolts out, so I had to pay a local repair shop to swap the factory shocks for the KYBs. Fantastic improvement in the ride. You can search Youtube for instructions on how to do this.

So the question goes back to OP.
If from the day one you never liked the softness/hardness -> check tire pressure -> change to softer tires -> change the shocks/struts.
If something went bad recently: Check air pressure -> have the shocks/struts examined. As I am getting introduced to third gen model, I have noticed that the original shocks tend to leak prematurely.
