All SC400s are auto.
Some SC300s have a 5 speed manual.

400 is a 4 liter V8, 300 is a 3 liter inline 6.
I don't know anything about swapping in a manual, I'll let someone else answer that.

400's never came with the manual transmission...only 92-97 SC300's as an option....

but search the forums, I don't think I've seen a 400 stick shift conversion yet....

from what i've heard, some people from australia converted their sc400s to manual, but im guessing it'll cost lots of money......and i also heard you'll need to fabricate a custom bellhousing to fit.....someone clarify me please

It would be cheaper to buy an sc300 manual and drop the v8 in it.

But then you might as well drop in a GTE (Supra motor for anyone new).

A new bell housing would be in order, as would changing the computer, because a single computer controls both the tranny and engine. The electronics is where most of the problems are and would be a much tougher and more expnsive.time consuming project.

I don't see the possibility of mating a manual transmission to a Lexus V8 without extensive and complicated modifications (even engineering changes).

Think about it, Lexus themselves never went through the trouble of putting a manual in their V8's.
If it wasn't that difficult and expensive, Lexus would of done it a long time ago even at a low sales rate.
