2001 ES 300 PCV Valve???
take off the Lexus plastic cover. PCV valve should be on top, back left corner of the intake manifold if you are standing in front.
Look for a black tubing clamped onto a black peice of plastic tubing that has a 90° angle to it. On end in the tubing, the other end goes into the manifold.
Search for PCV valve replacement and extend the search to cover threads up to a year. There was a thread a while ago that has very nice picture of the location and good/bad PCV valves.

I appreciate all the replies and pictures , but for the record, my 2001 ES engine is not built like the engine in the picture. I have no PCV at the same location. I have tentatively found that some engines may have been retro-fitted with a threaded brass PCV valve in the left rear (facing engine) valve cover. This may have been done to help with ventilation to mitigate the gelling problem with other V-6 engines.
