It's quite likely that you will have to remove that entire side panel in the rear to access the speaker.
One of the first things I did after buying a Lexus was to go to Harbor Freight and buy the set of blue plastic panel removers. There's 5-6 of them, cost is about $8.00 and they look like a bunch of odd-shaped plastic pry bars. I have found several of them very useful in removing all those pesky black clips under our hoods, and also in removing body panels without scratching them.
As an option (not a good one), Lexus sells ONE panel remover for about $45. I see no difference between it (yes, I was dumb enough to buy one) and the ones from Harbor Freight.
